Profit is measured by Taking the Total amount of Money Taken in, and then subtracting from it, the total amount of money spent.

Non for Profit do not measure profit, as they are not in it to make money.

In accounting we like to call them "not-for-profits" because that is more indicative of their business status. All other businesses are "for proft"- they are in it for the money, and absent making money would not exist. "Not-for-profits" may make money, but they are not in business for the purpose of making money. They are in business for some higher purpose. As such, a not-for-profit entity measures it's success by an increase in assets, not an increase in profits.

Every business measures it's profits. They do so with a Profit and Loss statement which, described very basically, lists the assets, lists all the expenditures then shows the difference between the two. All businesses hope that the assets will be more than the expenditures and therefore end up with a yearly "profit".

A non-profit business does a Profit and Loss Statement and they also hope to have a profit (even if it's a very small amount)because the only business that can run at a loss year after year and survive is the government. Non-profit is often associated with charities but many service businesses like membership organizations can also be non-profit. This means that the organization runs on a balanced budget each year.
