There will be Franchises of Share Broker Office in your area. Contact one of these agencies.

Also Indian Bank, HDFC Bank, ICICI Bank are also doing this business. Contact the nearest branch of the above banks

First you must open Depository Participant Account (DP Account)
followed by Trading Account with any share broker or with any of the above mentioned banks.

For this you require to fill necessary forms available with them and
you must produce documentary proof of your residence (set of documents approved for this purpose is available with these agencies)

You must also give a xerox copy of PAN Card of Income Tax Department.

When your DP Account and Trading Accounts are opened you can
start buying the shares on line.

Get an account with etrade, ameritrade, schwab or any other brokerage, fund your account with some amount of cash and go through their menus to buy and sell stocks.
If you have never traded stocks, it is best that you "paper trade" for a while to learn how stocks move. finance.yahoo.com has a perfect stock portfolio tracker that allows you to set up any number of test portfolios for free and then watch them move during the trading day. finance.google.com is an excellent place to get data on stocks and charts.
